I've been hanging around high-sec for a while now, and I'm trying to figure out when I should, or should have, been comfortable to venture into not so high sec. I know in essence it's different for each, but what are/were your markers, or what should have been your markers for being ready to go?

Well as RoKera touched on 0.0 I'll speak for low-sec (my opinion anyway!).
Personally I'd say just go for it. Yup skills, fittings and an income source behind you is all very important to have in place but so is getting some experience and learning through your own mistakes.
You don't really mention WHY you want to go into low or null though. Do you want to start pvp-ing? Do you just want to run through and have a look? Are you wanting to try pve like missions or probing in these areas?
If its just a matter of traveling around in low-sec then you will find you are relatively safe warping to 0 in a shuttle, pod or ideally an interceptor. Alternatively a cloaky ship. You will need to watch out for smart-bombers and it can be worth a look at the map for "pods destoryed in last hour" for any hot-spots that might be worth avoiding.
If you want to engage in pve activity well this is a little more tricky, you will need to be "pvp minded" in the sense of at least knowing how to avoid it if you are not fit out for it. You might need to adjust your normal pve fittings also. It really depends on what exactly you want to do.
As for pvp - there are some excellent guides out there on getting started with piracy and equally lots of good pirate corps who will train up new members. - I don't have any links to hand right now but asking on the Eve-O forums will probably be a good start. Alternatively there is Faction War - I'm a Faction War pilot myself and thats how I ended up living in low-sec. In a nutshell its one massive war-dec, so everything you are shooting is usually going to be a "legal" target, so no gate guns or sec-status loss. “a lot of it comes down to just biting the bullet and giving it a go” — definitely THIS. Just keep in mind “don’t fly what you can’t afford to lose,” and give it a shot. If everything goes well, great! If not, you can pull back and regroup, hopefully having learned a few things.
